Flow cytometric analysis of Mouse IL-7Rα (CD127) expression on BALB/c mouse splenocytes. BALB/c mouse splenocytes were stained with Brilliant Violet 421™ Rat Anti-Mouse CD3 antibody and either Biotin Rat IgG2a, κ Isotype Control (Left panel) or SDT Biotin Rat Anti-Mouse IL-7Rα (CD127) Antibody (Right panel) at 5 μl/test followed by Sav-PE. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
Interleukin-7 receptor α chain (IL-7Rα, CD127) is a 75 kDa type-I transmembrane glycoprotein that forms the cytokine-binding subunit of both the IL-7 receptor (IL-7R) and the thymic stromal lymphopoietin receptor (TSLPR); upon binding IL-7 it heterodimerizes with the common gamma chain (γc, CD132) to initiate JAK1/JAK3-mediated phosphorylation of STAT5, thereby driving survival, proliferation and differentiation of T, B and innate lymphoid cells, while TSLP engagement pairs IL-7Rα with the TSLPR chain to modulate type-2 immunity.
