Background

Perforin is a crucial protein involved in the immune response, primarily produced by cytotoxic T lymphocytes and natural killer cells. It functions as a pore-forming protein, inserting itself into the membranes of target cells, such as virus-infected or cancerous cells. Once integrated, perforin creates pores that disrupt the cell membrane's integrity, leading to osmotic lysis and ultimately killing the target cell. This mechanism is essential for the immune system's ability to eliminate harmful cells and maintain homeostasis.

Flow cytometric analysis of Perforin expression in PBMC (Human peripheral blood mononuclear cells). Human PBMC were fixed and permeabilized with Foxp3 / Transcription Factor Staining Buffer Set. The cells were then stained with Alexa Fluor® 647 Mouse Anti-Human CD4 Antibody and either Biotin Mouse IgG2b, κ Isotype Control or SDT Biotin Mouse Anti-Human Perforin Antibody at 5μl/test followed by Sav-iFluor 488. Total viable cells, as determined by Fixable Viability Dye 452 (S0D0021), were used for analysis. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
