Flow cytometric analysis of Human Granzyme B expression in Human PBMC. Human PBMC were fixed with 4% PFA and permeabilized with 0.1% Tween. The cells were stained with PE Mouse Anti-Human CD8 Antibody and either Biotin Mouse IgG1, κ Isotype Control (Left panel) or Biotin Mouse Anti-Human Granzyme B Antibody (Right panel) at 5 μl/test followed by Sav-iFluor 488. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
Granzyme B is a 32-kDa serine protease stored in the cytolytic granules of cytotoxic T lymphocytes and natural killer cells, whose immune function centers on perforin-assisted translocation into target cells where it directly cleaves and activates pro-apoptotic substrates such as procaspase-3, Bid and PARP, thereby igniting both caspase-dependent and caspase-independent mitochondrial apoptosis pathways to eliminate virus-infected or malignant cells.
