Flow cytometric analysis of Mouse CD9 expression on C57BL/6 mouse bone marrow. C57BL/6 mouse bone marrow was stained with APC-Cy7 Rat Anti-Mouse Ly-6G/Ly-6C Antibody and either Alexa Fluor® 488 Mouse Ig2a, κ Isotype Control (Left panel) or SDT Alexa Fluor® 488 Rat Anti-Mouse CD9 Antibody (Right panel) at 1.25μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD9 is a 24-kDa tetraspanin membrane glycoprotein with four transmembrane domains and two extracellular loops that is widely expressed on hematopoietic and non-hematopoietic cells, where it organizes tetraspanin-enriched microdomains that integrate adhesion, signaling and trafficking by associating with integrins, EWI-2/F, and other tetraspanins to regulate cell adhesion, migration, fusion, exosome biogenesis/secretion, immune activation and tumor suppression, and its large extracellular loop is essential for sperm–egg fusion, while its presence in exosomes marks these vesicles for intercellular communication and its altered expression influences cancer progression, viral entry and stem-cell pluripotency.
