Background
IL-37b (Interleukin-37 isoform b, also known as IL-1F7b, is an anti-inflammatory cytokine in the IL-1 family. It contains a β-trefoil fold composed of 12 β-strands and is the largest and most extensively studied isoform of IL-37. Through the IL-18Rα/IL-1R8 receptor complex and a SMAD3-dependent pathway, it inhibits the production of pro-inflammatory cytokines such as IL-1α and IL-6, and downregulates signaling pathways including NF-κB and PI3K-Akt. Clinical studies have found that IL-37b is abnormally expressed in inflammatory diseases such as rheumatoid arthritis, inflammatory bowel disease, and allergic asthma; mouse models show that it has protective effects and is a potential therapeutic target for inflammatory diseases.
