Background

NRG1β1 (also known as HRG1-β1, heregulin beta-1) is a soluble isoform of Neuregulin-1 (NRG1) generated by alternative splicing. The NRG1 gene is located on human chromosome 8p12 and belongs to the NRG gene family (NRG1, NRG2, NRG3, NRG4).

NRG1β1 contains an immunoglobulin (Ig)-like domain and an epidermal growth factor (EGF)-like domain. The EGF-like domain, encoded by exons 6 and 7, is essential for its biological activity and receptor binding. Extensive alternative splicing of the NRG1 gene generates α and β variants at the C-terminal region of the EGF-like domain, with isoforms containing the β-variant EGF-like domain exhibiting significantly higher binding affinity for ErbB3 or ErbB4 homodimers than α-variant isoforms.
