Background

Growth hormone (GH), also called somatotropin, is a polypeptide hormone secreted mainly by the somatotrophs of the anterior pituitary gland. Growth hormone plays a major role in postnatal growth but is also essential for metabolic regulation. It is mainly used clinically for the classification of pituitary tumors and the study of neuroendocrine tumors.

Western Blot

WB result of Human Growth Hormone / GH Recombinant Rabbit mAb
Primary antibody: Human Growth Hormone / GH Recombinant Rabbit mAb at 1/1000 dilution
Lane 1: mouse liver lysate 20 µg
Lane 2: mouse pituitary lysate 20 µg
Negative control: mouse liver lysate
Secondary antibody: Goat Anti-rabbit IgG, (H+L), HRP conjugated at 1/10000 dilution
Predicted MW: 25 kDa
Observed MW: 22 kDa

IP

Human Growth Hormone / GH Rabbit mAb at 1/50 dilution (1 µg) immunoprecipitating Human Growth Hormone / GH in 0.4 mg mouse pituitary lysate.
Western blot was performed on the immunoprecipitate using Human Growth Hormone / GH Rabbit mAb at 1/1000 dilution.
Secondary antibody (HRP) for IP was used at 1/1000 dilution.
Lane 1: mouse pituitary lysate 20 µg (Input)
Lane 2: Human Growth Hormone / GH Rabbit mAb IP in mouse pituitary lysate
Lane 3: Rabbit monoclonal IgG IP in mouse pituitary lysate
Predicted MW: 25 kDa
Observed MW: 22 kDa
This blot was developed with high sensitivity substrate

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human pituitary. Anti- Human Growth Hormone / GH antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

IHC shows positive staining in paraffin-embedded human placenta.

Negative control: IHC shows negative staining in paraffin-embedded human tonsil.

Negative control: IHC shows negative staining in paraffin-embedded human kidney.

Immunofluorescence

IF shows positive staining in paraffin-embedded human placenta. Anti-Human Growth Hormone / GH antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
