Background

YTHDF1 (YTH N6-Methyladenosine RNA Binding Protein 1) is a cytoplasmic reader protein that specifically recognizes and binds to N6-methyladenosine (m6A), the most prevalent internal epitranscriptomic modification in eukaryotic messenger RNAs. Containing a conserved YTH (YT521-B homology) domain that mediates selective m6A recognition, YTHDF1 promotes translation efficiency of its target transcripts by facilitating ribosome loading and interacting with translation initiation factors, including eIF3 complex components. Through this mechanism, YTHDF1 enhances the expression of critical oncogenes and cell cycle regulators, thereby influencing cell proliferation, stem cell maintenance, and immune responses. Notably, YTHDF1 has emerged as a significant player in cancer biology, where its overexpression in various malignancies—including hepatocellular carcinoma, gastric cancer, and acute myeloid leukemia—correlates with tumor progression, metastasis, and poor clinical outcomes. Additionally, YTHDF1-mediated m6A recognition regulates antitumor immunity by modulating the translation of immune checkpoint molecules and cytokine transcripts in the tumor microenvironment. Beyond cancer, YTHDF1 contributes to synaptic plasticity and learning memory in the nervous system, as well as to viral replication through selective enhancement of viral mRNA translation. The therapeutic potential of targeting YTHDF1-m6A interactions is currently being explored through the development of small molecule inhibitors that disrupt this oncogenic axis.

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-YTHDF1 antibody was used at 1/500 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
