Background

Villin is a cytoskeletal protein with a molecular weight of 95kDa, which is an actin binding protein. It exists in the cytoskeleton of microvilli and plays a crucial role in maintaining the brush like edge tissue structure. Normally distributed in the intestinal epithelium and renal proximal tubule epithelium, it can be used for the differential diagnosis of intestinal epithelial derived tumors and non-intestinal epithelial tumors, and can also serve as a reference indicator for the diagnosis of gastrointestinal neuroendocrine tumors.

Immunocytochemistry

ICC shows positive staining in HT-29 cells. Anti- Villin antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (red).
