Background

Versican is a large extracellular matrix proteoglycan primarily secreted by mesenchymal cells and belonging to the hyaluronan-binding lectican family; its core structure comprises an N-terminal G1 domain (responsible for stable binding to hyaluronan and link proteins), a central region containing glycosaminoglycan (GAG) attachment domains (GAG-β and/or GAG-α) that are covalently linked to chondroitin sulfate chains to confer significant hydration capacity, turgor pressure, and elastic buffering to tissues, and a C-terminal G3 domain that mediates interactions with other matrix components such as fibulins, selectins, and growth factors. This protein is critical during embryonic development for heart formation, neural crest cell migration, and limb morphogenesis, while in adult tissues it plays a widespread role in vascular homeostasis, wound healing, and the regulation of inflammatory responses; moreover, its aberrant overexpression is closely associated with the remodeling of the tumor microenvironment, where it promotes cancer cell proliferation, migration, invasion, and immune evasion, establishing Versican as a key biomarker for the progression and poor prognosis of various malignant tumors.
