Background

VDAC3 (Voltage-Dependent Anion Channel 3) is a crucial isoform of the mitochondrial outer membrane porin family that distinguishes itself from its counterparts, VDAC1 and VDAC2, through unique structural features including an additional cysteine residue which facilitates the formation of intermolecular disulfide bonds and potential oligomerization. Primarily localized in the mitochondria of diverse tissues with particularly high expression in the brain, testes, and skeletal muscle, VDAC3 plays a specialized role in regulating metabolite flux, such as ATP and ADP exchange, while critically contributing to the assembly and stability of the sperm flagellar axoneme, where its dysfunction is directly linked to male infertility due to impaired sperm motility. Beyond its metabolic gating function, VDAC3 is implicated in modulating reactive oxygen species (ROS) levels and interacting with cytoskeletal elements, suggesting a broader involvement in cellular signaling pathways related to oxidative stress response and apoptosis, although its precise gating mechanisms and specific protein-protein interaction networks remain less fully characterized compared to the more ubiquitous VDAC1.
