Background

B-cell maturation antigen (BCMA, gene name TNFRSF17) is a member of the tumor necrosis factor receptor superfamily (TNFRSF) that is specifically expressed on late-stage B-cell lineage cells, including plasmablasts and long-lived plasma cells, while being virtually absent in normal tissues or early B-cells. As a key receptor for both BAFF (B-cell activating factor) and APRIL (a proliferation-inducing ligand), BCMA recruits TRAF adapter proteins to activate downstream signaling pathways such as NF-κB, MAPK, and PI3K/Akt, playing a central role in sustaining plasma cell survival, differentiation, and antibody secretion. Due to its highly restricted expression profile and its aberrant overexpression on multiple myeloma (MM) cells, BCMA has emerged as a premier therapeutic target; consequently, various BCMA-directed therapies, including chimeric antigen receptor T-cell (CAR-T) therapy, bispecific antibodies, and antibody-drug conjugates (ADCs), have demonstrated remarkable clinical efficacy, fundamentally transforming the treatment landscape for this disease.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in U266B1 cells (top panel) and negative staining in HeLa cells (below panel). Anti- TNFRSF17/BCMA antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
