Background

TAOK1 (TAO kinase 1) is a multifunctional serine/threonine protein kinase and a member of the MAP3K family. It activates the p38/MAPK14 stress signaling pathway through phosphorylation of MAP2K3/MAP2K6, regulates microtubule dynamic stability by phosphorylating MARK2 (its ability to phosphorylate TAU protein is associated with tauopathies), and participates in DNA damage checkpoints as well as the JNK apoptotic cascade. In the nervous system, TAOK1 induces neurite initiation via phosphorylation of MARK2 and downstream TAU; its autophosphorylation at Thr440 or phosphorylation by MST3 kinase enables interaction with myosin Va to promote dendritic spine and synaptic maturation. Mutations in the TAOK1 gene have been confirmed to be closely associated with autosomal dominant neurodevelopmental disorders, including developmental delay, autism spectrum disorder, and intellectual disability. Furthermore, in cardiomyocytes, this kinase alleviates heart failure-related damage by inhibiting NLRP3 inflammasome activation and restoring YAP/TEAD pathway activity.
