Background
Gulgafafusp alfa is a humanized IgG2κ monoclonal antibody fusion protein targeting the glucagon-like peptide-1 receptor (GLP-1R) for the treatment of type 2 diabetes mellitus and obesity. This protein is engineered through genetic fusion of a GLP-1(7-35) agonist fragment to the Fc region of an anti-GLP-1R antibody, forming a unique "antibody-ligand" bifunctional structure: the antibody moiety specifically binds to GLP-1R, while the GLP-1 fragment activates downstream cAMP signaling pathways. Through synergistic dual receptor binding sites, this design significantly enhances binding affinity and prolongs half-life.
