Background

AGTR1, the angiotensin II type 1 receptor, is a seven-transmembrane G-protein-coupled receptor that serves as the principal transducer of angiotensin II signaling in the renin–angiotensin–aldosterone system, mediating potent vasoconstriction, aldosterone and vasopressin release, renal sodium and water reabsorption, vascular smooth-muscle and cardiac hypertrophy, oxidative stress generation and sympathetic activation, thereby critically regulating systemic blood pressure and cardiovascular homeostasis; the receptor is widely expressed in kidney, heart, vasculature, adrenal gland and lung, can be activated not only by ligand binding but also by mechanical stretch, is transcriptionally up-regulated by inflammatory cytokines, undergoes endocytosis and heterodimerization that fine-tune its signaling output, and its chronic overactivity drives hypertension, cardiac and renal fibrosis, atherosclerosis and diabetic complications, while genetic polymorphisms that increase its expression or alter its regulation are linked to salt-sensitive hypertension and heightened cardiovascular risk .

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-AGTR1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
