Background
α-Actinin-4 (ACTN4) is a widely expressed non-muscle F-actin crosslinking protein belonging to the α-actinin family, encoded by the ACTN4 gene located on human chromosome 19q13, containing approximately 911 amino acids with a molecular weight of about 104.9 kDa. Its structure consists of an N-terminal actin-binding domain (ABD) composed of two calponin homology domains (CH1 and CH2), a central rod domain formed by spectrin repeats, and a C-terminal EF-hand calcium-binding motif, and it exists as an antiparallel dumbbell-shaped homodimer. ACTN4 is highly expressed in glomerular podocytes and is essential for the architecture and adhesion of podocyte foot processes. Mutations in its gene can cause autosomal dominant focal segmental glomerulosclerosis (FSGS); all identified pathogenic mutations are point mutations within the ABD (such as K255E and I150M), which increase the protein's affinity for F-actin by exposing a hidden actin-binding site, while the mutant protein is also more susceptible to degradation via the ubiquitin-proteasome pathway, thereby exhibiting both gain-of-function and loss-of-function effects that lead to podocyte dysfunction and proteinuria. In addition, ACTN4 is highly expressed in various malignant tumors and is associated with tumor invasion, metastasis, and poor prognosis, serving as a biomarker for malignant cell invasion and drug resistance.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti-α-Actinin-4 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
