Background
53BP1 (p53-binding protein 1) is a large, multi-domain scaffold protein that rapidly accumulates in irradiation-induced foci at DNA double-strand breaks, where it acts as a master regulator of the non-homologous end-joining repair pathway by binding phosphorylated H2AX (γH2AX) and histone H4 methylated on Lys20, thereby recruiting the downstream effector kinase ATM, the ubiquitin ligases RNF8/RNF168, and the adaptor proteins BRCA1 and RIF1 to antagonize BRCA1-mediated DNA-end resection and channel repair toward the error-prone but fast NHEJ route, while its own recruitment and retention are governed by tandem Tudor and BRCT domains that sense combinatorial histone marks, and its loss or mutation leads to genomic instability, hypersensitivity to ionizing radiation, and impaired class-switch recombination in B lymphocytes.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
