Theory This kit adopts double antibody sandwich enzyme-linked immunosorbent detection technology. Specific anti-human IL-12 p70 antibodies were pre-coated on high affinity labeled plates. The standard substance, the sample to be tested and the biotinylated detection antibody are added to the well of the enzyme label plate. After incubation, IL-12 p70 present in the sample binds to the solid phase antibody and the detection antibody to form an immune complex. After washing to remove unbound material, horseradish peroxidase-labeled Streptavidin-HRP was added. After washing, a chromogenic substrate is added to protect the color from light. A stop solution was added to stop the reaction, and the absorbance value was measured at a wavelength of 450 nm (reference calibration wavelength of 540 nm or 570 nm).

Background Interleukin 12 p70 (IL-12), also known as natural killer cell stimulating factor (NKSF) or cytotoxic lymphocyte maturation factor (CLMF), is a heterodimeric pleiotropic cytokine consisting of a 40kDa (p40) subunit and a 35kDa (p35) subunit. Human p40 may circulate as a monomer, a homodimer, or a complex that binds to p35 to form IL-12 or binds to p19 to form IL-23.
Both homodimers p40 and IL-23 can bind to IL-12R as non-signal antagonists. Alternatively, p40 can also bind to Rβ1, activating microglia and macrophages. The IL-12 p40 subunit shares with another heterodimeric cytokine, IL-23, with similar and distinct biological activity from IL-12. Cells known to produce IL-12 include macrophages and dendritic cells, monocytes, Langerhans cells, neutrophils, keratinocytes, plasmacytoid dendritic cells, microglia, CD8 + DCs (mouse cells only), and non-germinal center (CD38-CD44 +) B cells (human cells only). Functional IL-12 was shown to enhance cytotoxic activity and induce interferon gamma (IFN-gamma) production within NK cells, T cells, and dendritic epidermal T cells. IL12 has also been reported to induce IFN-γ production by macrophages. IL-12, together with other members of the IL-12 family, IL-23 and IL-27, promotes the development of CD4+Th1 immune response. In response to infection, IL-27 is first released, promoting the transition from Th0 to Th0/1. IL-12 is subsequently produced, resulting in Th1 effector cells. IL-12 combines with IL-18 to generate Th1 memory cells from effector cells, which are subsequently activated by IL-23. While mouse IL-12 is active on both human and mouse cells, human IL-12 is not active on mouse cells.
