Theory

Double antibody sandwich enzyme-linked immunosorbent assay was used in this kit. Specific anti-human CXCL10/IP-10 antibody was precoated on a high-affinity microplate. After incubation, CXCL10/IP-10 in the sample was combined with the solid-phase antibody and the detection antibody to form an immune complex. After washing to remove the unbound material, the horseradish peroxidase labeled Streptavidin-HRP was added. After washing, the chromogenic substrate was added and the color was developed in the dark. The reaction was terminated by adding the termination solution, and the absorbance value was measured at 450nm wavelength (reference correction wavelength 540nm or 570nm).

Background

Chemokine ligand 10 (CXCL10), also known as interferon-inducible protein 10 (IP-10) or micro-inducible cytokine B10, is a small molecular weight cytokine belonging to the CXC chemokine family. Many types of cells can secrete IP-10 in response to IFN-γ, including monocytes, endothelial cells and fibroblasts. IP - 10 has a variety of functions, such as the monocyte/macrophage, T cells, NK cells and dendritic cell chemotaxis, promote T cell adhesion to endothelial cells, antitumor activity and inhibition of bone marrow colony forming and angiogenesis.
