NLRP3 is a Member of the NLR Family
NLRP3 is a member of the NLR (Nod-like Receptor) family and a key component of the NLRP3 inflammasome. It can activate caspase-1, which in turn cleaves and matures IL-1β (interleukin-1β) and IL-18 (interleukin-18). It is important to note that the NLRP3 inflammasome only assembles after being stimulated by various microbial stimuli and endogenous stimuli; therefore, NLRP3 does not directly interact with its activators. NLRP3 is highly expressed in dendritic cells, monocytes, and macrophages.
When no bands appear in Western Blotting (WB), the first step is to rule out issues related to the sample's expression level and confirm the sample's expression level.
Try treating the sample with LPS (Lipopolysaccharide) or PGN (Peptidoglycan).
Perform ultrasonic treatment to fully disrupt the cell membrane, nuclear membrane, and chromatin. This enhances the homogeneity of the sample, increases protein yield, and reduces background noise at the same time.Experimental Procedures
