Background
Tumor necrosis factor α (TNF alpha) is an effective pro-inflammatory cytokine, which can kill and inhibit tumor cells. It is mainly produced by activated macrophages and can also be secreted by other types of cells, such as CD4+ lymphocytes, NK cells, neutrophils, mast cells, eosinophils and neuronal tissues. The members of TNF alpha family exert their cellular effect through two distinct surface receptors of the TNF receptor family, TNFR1and TNFR2. The pleiotropic biological effects of TNF can be attributed to its ability to simultaneously activate multiple signaling pathways in cells. TNF-induced NF-κB activation promotes the growth, invasion and metastasis of cancer cells.

Linearity
The concentrations of Mouse TNF-α were measured and interpolated from the target standard curves and corrected for sample dilution.
The sample is undiluted samples are as follows: RAW264.7 mouse monocyte/macrophage cells stimulated with 5ug/ml LPS and 100ng/mL rmIL-10 for 7days (5%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 5603.07 pg/mL in stimulated RAW264.7 supernatant.
Linearity
The sample is undiluted samples are as follows: RAW264.7 mouse monocyte/macrophage cells culture for 7days (100%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be205.69 pg/mL in unstimulated RAW264.7 cells supernatant.
To assess the linearity of the assay, sample was spiked with high concentrations of Mouse TNF-α in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
