Background
Mouse Interleukin-10 (IL-10) is a potent, pleiotropic cytokine that plays a critical role as a central regulator of immune responses; it is a homodimeric protein produced by a wide variety of immune cells, including macrophages, dendritic cells, and T and B lymphocytes, and its primary biological function is to limit and ultimately terminate inflammatory responses by inhibiting the expression of pro-inflammatory cytokines (such as TNF-α, IL-1, and IL-6) and chemokines from activated macrophages and dendritic cells, while also downregulating major histocompatibility complex (MHC) class II and co-stimulatory molecules to suppress antigen presentation and T-cell activation. By controlling the magnitude and duration of inflammation, Mouse IL-10 is essential for protecting the host from immunopathology during infections and autoimmune reactions, though its potent anti-inflammatory activity can also be exploited by pathogens to evade the immune system; due to its central role in immune homeostasis, the IL-10 gene knockout mouse is a valuable model for studying inflammatory bowel disease (IBD) and other chronic inflammatory conditions.

ELISA
To assess the linearity of the assay, three samples were spiked with high concentrations of Mouse IL-10 in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
