This difference causes exon 7 to be extensively skipped during the splicing of SMN2 pre-mRNA, with approximately 90% of transcripts producing a truncated protein lacking exon 7 (SMNΔ7), which is unstable and functionally impaired. As a result, SMN2 can only produce a small amount of functional full-length SMN protein, which is insufficient to fully compensate for the protein deficiency caused by SMN1 loss.
