The catalytic mechanism of SCD depends on a di-iron center coordinated by nine conserved histidine residues, which activates molecular oxygen, and it also requires cytochrome b5 as an electron donor to regenerate the iron center. Structural biology studies have revealed that SCD1 adopts a unique fold: four transmembrane helices form a conical structure capped by a cytosolic domain, with the substrate acyl chain enclosed within a narrow tunnel approximately 24 Å long that contains a sharp bend at the C9 and C10 positions, thereby precisely determining the positional selectivity and stereospecificity of the desaturation reaction. Functionally, SCD1 knockout mice exhibit a lean, hypermetabolic, insulin-sensitive phenotype with resistance to diet-induced obesity, making SCD an attractive drug target for metabolic diseases such as obesity, diabetes, and non-alcoholic fatty liver disease.
