As the functional equivalent of Escherichia coli L10 protein, RPLP0 belongs to the L10P family, and its C-terminus is highly similar to the acidic ribosomal phosphoproteins P1 and P2. By binding to P1\/P2 dimers, it forms a pentameric complex composed of a P1₂-P2₂ tetramer and a P0 monomer; this structure is essential for GTP hydrolysis during translation and for the recruitment of translation factors such as eEF2. RPLP0 is widely expressed in normal tissues, but recent studies have revealed that it is significantly upregulated in various tumors including clear cell renal cell carcinoma and lung adenocarcinoma, where high expression is independently associated with advanced stage, metastasis, and poor prognosis; its mechanisms involve promoting epithelial-mesenchymal transition (EMT), regulating the cell cycle, and influencing the immune microenvironment. Notably, RPLP0 also has a nuclear function independent of the ribosome: DNA damage-induced paraspeckle formation can recruit RPLP0 to bind the NONO protein, enhancing DNA-PK autophosphorylation and thereby promoting non-homologous end joining repair and tumor radioresistance.
