The human PRTN3 gene is located at chromosome 19p13.3, clustered with the neutrophil elastase (ELANE) and azurocidin genes; its expression is largely restricted to the promyelocyte\/myelocyte stage of bone marrow development and is regulated by G-CSF and the PU.1 transcription factor. In terms of cellular localization, PRTN3 is stored mainly in the azurophilic granules of neutrophils and is released upon stimulation of neutrophils by TNF, LPS, or fMLP; after release, it can anchor to the cell surface by binding to CD177. Functionally, PRTN3 degrades elastin, fibronectin, laminin, and type I, III, and IV collagen, and enhances endothelial barrier function by cleaving and activating PAR-2, participating in the regulation of neutrophil transendothelial migration. Clinically, PRTN3 is one of the principal autoantigens in antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis, and anti-PR3 antibodies (PR3-ANCA) are closely associated with the diagnosis, disease activity, and relapse risk of granulomatosis with polyangiitis. Recent studies have also found that serum PRTN3 levels can predict the prognosis of patients with metastatic colorectal cancer receiving first-line chemotherapy (especially bevacizumab treatment), with high PRTN3 levels associated with worse progression-free survival; the mechanism may involve PRTN3 promoting angiogenesis and attenuating the antiangiogenic effect of bevacizumab. In addition, PRTN3 has attracted attention in myeloid hematopoietic differentiation and acute myeloid leukemia, suggesting that its functions extend well beyond serving as an autoantigen in vasculitis.\u003c\/span\u003e\u003c\/p\u003e\u003c\/div\u003e","brand":"Starter","offers":[{"title":"25μl","offer_id":67399170621515,"sku":"S0B60773-25μl","price":100.0,"currency_code":"USD","in_stock":true},{"title":"100μl","offer_id":67399170654283,"sku":"S0B60773-100μl","price":350.0,"currency_code":"USD","in_stock":true}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0590\/8375\/1499\/files\/d82618d1c4f84406ae90db181ca4eb55_dbb0ce0a-a841-4063-9429-488018cc8159.jpg?v=1793847624","url":"https:\/\/www.antbioinc.com\/products\/prtn3-recombinant-rabbit-mab-s-4942-65-s0b60773","provider":"AntBio","version":"1.0","type":"link"}
