Human PRODH2 is 536 amino acids long, with its catalytic core located between residues 157 and 515; it strongly prefers hydroxyproline over proline as a substrate and uses coenzyme Q10 rather than oxygen as the terminal electron acceptor in vivo, and is therefore fundamentally a dehydrogenase rather than an oxidase. The enzyme is expressed at highest levels in the kidney and liver, and its downstream product glyoxylate can lead to elevated oxalate and calcium oxalate kidney stone formation in individuals with oxalate metabolism disorders, so PRODH2 is regarded as a potential therapeutic target for primary hyperoxaluria. In terms of genetic disease, biallelic pathogenic variants in PRODH2 are the genetic basis of hydroxyprolinemia, an autosomal recessive benign biochemical phenotype; patients are usually asymptomatic, but because hydroxyproline is difficult to distinguish from isoleucine\/leucine in tandem mass spectrometry, it often causes false-positive newborn screening for maple syrup urine disease. In addition, recent studies have found that PRODH2 overexpression can significantly enhance the in vivo antitumor efficacy of CAR-T cells, expanding the potential application value of this metabolic enzyme in immunotherapy engineering.\u003c\/span\u003e\u003c\/p\u003e\u003c\/div\u003e","brand":"Starter","offers":[{"title":"25μl","offer_id":67399173963851,"sku":"S0B60772-25μl","price":100.0,"currency_code":"USD","in_stock":true},{"title":"100μl","offer_id":67399173996619,"sku":"S0B60772-100μl","price":350.0,"currency_code":"USD","in_stock":true}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0590\/8375\/1499\/files\/76cf6da263274c2fb10c14ac62c9d87a_885c83d7-f652-4194-9828-a3a33ae0d1ce.jpg?v=1793847628","url":"https:\/\/www.antbioinc.com\/products\/prodh2-recombinant-rabbit-mab-s-5085-19-s0b60772","provider":"AntBio","version":"1.0","type":"link"}
