However, in various neurodegenerative diseases such as amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration (FTLD), TDP-43 mislocalizes from the nucleus to the cytoplasm and aggregates to form insoluble, hyperphosphorylated inclusions, with phosphorylation at Ser409\/410 being a core feature of these pathological inclusions. This phosphorylation modification is considered to be closely associated with disease progression—it not only serves as a \"molecular fingerprint\" of pathological conformational changes in TDP-43 but may also promote its aggregation, exacerbate cytotoxicity, and interfere with the autophagy-lysosomal clearance pathway.
