Its classic function is to serve as a high-affinity receptor for proteinase 3 (PR3) on the neutrophil surface, forming the CD177–PR3 membrane complex that maintains high-density presentation of PR3 on the cell surface; this complex constitutes a key target for autoantibody attack in PR3-ANCA-associated vasculitis, and blocking the binding of CD177 to PR3 can significantly reduce ANCA-induced neutrophil respiratory burst. In addition, CD177 can participate in neutrophil transendothelial migration by binding PECAM-1 on endothelial cells, and may influence the balance between adhesion and migration by regulating β2 integrin signaling. Recent studies have further revealed that CD177 is also expressed on the surface of some tumor-infiltrating regulatory T cells and is associated with enhanced immunosuppressive activity and tumor immune evasion.
